Phenobanate, discovered and developed by SK Biopharmaceuticals and its U.S. subsidiary SK Life Sciences, is considered to be a sodium channel (sodium channel) blocker by inhibiting voltage-gated sodium currents and positively regulating GABAA (gamma-aminobutyric acid) receptor activity through isomerism. Phenobarate is used to treat partial-onset epilepsy in adults, but it is not yet known whether it is safe and effective in children. The drug is a prescription drug and became available in the United States in 2019. Epilepsy occurs due to the uncontrolled activation of a group of nerve cells in the brain. Partial-onset epilepsy refers to epileptic seizures originating in a limited part of the brain. Phenobarate, taken orally once a day, can treat partial-onset epilepsy in adults. It is an innovative treatment for epilepsy independently developed by SK Life Science.

Three clinical trials involving more than 1,900 patients showed significantly higher response rates for all doses of phenobarnate compared with placebo during the 12-week maintenance period. The response rates of the three groups were: 40% for 100mg/day (p=0.036); 56% for 200mg/day (p<0.001); 64% for 400mg/day (p<0.001); and 25% for the placebo group. During the maintenance period, 4%, 11%, and 21% of patients treated with phenobarnate experienced a 100% reduction in seizure frequency at three doses, respectively, compared with 1% in the placebo group. This trial confirms that phenobanamate is an effective treatment option for adults with focal-onset epilepsy whose condition is uncontrolled on one to three antiepileptic drugs (ASMs).
