Introduction: Stipentol is a prescription drug used to treat epileptic seizures associated with Dravet syndrome. Dravet syndrome, also known as severe myoclonic epilepsy in infants, is a rare clinically refractory epilepsy syndrome in children. Anti-seizure drugs usually have poor efficacy. The main purpose of treatment is to reduce the frequency of seizures and reduce the occurrence of status epilepticus.

Indications for stiripentol

It is indicated for the treatment of seizures associated with Dravet syndrome in patients 6 months or older, weighing 15 pounds or more, taking clobazam. There are no clinical data to support the use of stiripentol (Diacomit) alone in the treatment of Dravet syndrome. The original developer of stiripentol is the French company Biocodex, which is available in two dosage forms: capsules and dry suspensions. Currently, stiripentol dry suspension is imitated by Shijiazhuang No. 4 Pharmaceutical Co., Ltd. in China.

Principle of action

Stipentol is a gamma-aminobutyric acid (GABA) enhancer. It mainly works by directly acting on the neurotransmitter gamma-aminobutyric acid (GABA) receptors to enhance GABAergic nerve conduction. It also inhibits a variety of liver cytochrome P450 enzymes and increases the blood concentration of some anti-epileptic drugs, thereby exerting anti-epileptic effects.

Efficacy of stiripentol in the treatment of epilepsy

Stipentol has clinically demonstrated efficacy in the treatment of seizures associated with Dravet syndrome and has been shown to control clonic and tonic-clonic seizures.

Precautions for stiripentol

1. Drowsiness: Stipentol can cause drowsiness. It is recommended to monitor drowsiness during medication. Especially when stiripentol is used with other inhibitors or clobazam, try not to engage in driving operations or fine work.

2. Loss of appetite and weight loss: Stipentol can cause loss of appetite and weight loss. Pay attention to monitoring weight changes.

3. Changes in blood cell counts: Stipentol can cause a significant decrease in white blood cell and platelet counts, so blood tests should be performed before starting stiripentol treatment and then every 6 months.

4. Withdrawal symptoms: Like most anti-epileptic drugs, stiripentol should be discontinued gradually to reduce the risk of higher frequency of epileptic seizures and status epilepticus. The drug should not be stopped suddenly.
