However, patients who require an urgent increase in serum sodium to prevent or treat severe neurological symptoms should not be treated with tolvaptan tablets.
In a phase III clinical trial, one group was given tolvaptan 7.5 mg/d, and the other group was given placebo for 7 days. Both were used on the basis of traditional treatment. The results showed that the improvement rate of lower limb edema in the tolvaptan group was also significantly higher than that of the placebo group, 54.8% vs. 28.3%, P=0.0168; there was also a significant difference in the changes in ascites volume between the two groups, placebo group vs. tolvaptan group (-191.8±690.8) mL vs. (-492.4±760.3) mL, P=0.0093. Changes in abdominal circumference were significantly better in the tolvaptan group than in the placebo group. This shows that tolvaptan is better than placebo in reducing weight, reducing ascites volume, and improving lower limb edema.
