Grapefruit juice slows the metabolism of this drug, which is regulated by CYP3A4, and should not be used to administer or dilute this drug. It is recommended to take sirolimus 4 hours after taking cyclosporine oral solution (modified version) and/or cyclosporine capsules (modified version).

Different conditions require different treatment courses. However, the drug has side effects, and any drug is not suitable for long-term use. Adverse reactions include side effects and toxic reactions. Side effects are inherent and unavoidable with drugs, but they are not harmful. Toxic effects are very harmful and can be avoided. Long-term use will cause toxic effects, so it is recommended that you do not take certain drugs, including rapamycin (sirolimus), for a long time.

Rapamin (sirolimus) is rapidly absorbed from oral solution, with the average time to peak after a single oral dose being approximately 1 hour; in kidney transplant recipients, the average time to peak after multiple oral doses is approximately 2 hours. High-fat meals can increase the absorption of rapamin (sirolimus), so it is recommended that oral rapamin (sirolimus) should be taken regularly with or without food.

Although the loading dose of 15 mg and the maintenance dose of 5 mg/day used in clinical trials are safe and effective, the efficacy benefit of doses above 2 mg in renal transplant patients is unclear. The overall safety profile of patients taking rapamycin (sirolimus) oral solution 2 mg daily was better than that of patients taking 5 mg oral solution daily.
