有关唑来膦酸的那些你不太了解的事
Bisphosphonate drugs can be divided into two categories: nitrogen-free and nitrogen-containing. After 20 years of development, early nitrogen-free bisphosphonate drugs, such as clodronate and etidronate, have developed to second and third generation nitrogen-containing bisphosphonate drugs, such as pamabisphosphonic acid, zoledronic acid, etc., and their efficacy has become increasingly stronger. Nitrogen-free bisphosphonates have a direct toxic effect on cells, thereby inducing cell apoptosis, while nitrogen-containing bisphosphonates inhibit the bone surface activity of cells by interfering with the information transmission between osteoclasts. Therefore, nitrogen-containing bisphosphonates have a stronger inhibitory effect on bone resorption. Today I’m going to tell you about some things you don’t know much about.
Zoledronic acid contains a unique dinitrogen structure, has a high affinity with bone tissue, and has a long action cycle. It can affect osteoclast absorption, differentiation and aggregation in a variety of ways, thereby inducing osteoclast death. Studies have shown that zoledronate sodium can effectively inhibit bone loss and improve bone density.
Zoledronic acid should be administered intravenously. For adults, 4 mg each time should be diluted with 100 ml of 0.9% sodium chloride injection or 5% glucose injection and then infused intravenously. The infusion time should be no less than 15 minutes. Administer every 3-4 weeks or as directed by your doctor. Its common side effects are excitement, headache, fatigue, conjunctivitis, hypomagnesemia, nausea, diarrhea, dysphagia, leg edema, hypokalemia, vomiting, constipation, hypophosphatemia, hypocalcemia, anorexia, hypotension, granulocytopenia, anemia, chest pain, pancytopenia, abdominal pain, bone pain, joint pain, thrombocytopenia, myalgia, insomnia, anxiety, drowsiness, etc.
