依普利酮效果怎样？

How effective is eplerenone? A clinical trial was conducted to explore the effects of eplerenone (Planep) and losartan on arterial stiffness and central arterial pressure (CAP) in patients with dual-stage mild to moderate essential hypertension who had elevated systolic and diastolic blood pressure.

The trial included 84 patients with mild to moderate essential hypertension [diastolic blood pressure 90-110mm Hg (1mm Hg=0.133kPa) and systolic blood pressure 140-180mm Hg] were divided into eplerenone group and losartan group, with 42 patients in each group. After taking placebo for 2 weeks, they entered the 12-week trial period. They were given eplerenone (Planep) 50 mg or losartan 50 mg orally once/d, followed by a follow-up visit once/2 weeks. After 4 weeks, if the diastolic blood pressure is ≥90 mm Hg, they will be changed to eplerenone 100 mg orally divided into 2 times or Losartan 100 mg orally once/d.

The test results showed that: 37 patients with hypertension in the eplerenone (Planep) group completed the trial and 5 were lost to follow-up; 40 patients in the losartan group completed the trial and 2 were lost to follow-up; compared with before treatment, the sitting systolic and diastolic blood pressure, ambulatory blood pressure parameters, and b aPWV and CAP were significantly reduced; at the end of 12 weeks of treatment, there was no significant difference in the decrease in sitting systolic blood pressure and diastolic blood pressure between the two groups [(18.3±8.6) vs. (21.4±9.4), (14.0±5.3) vs. (15.1±6.6) mm Hg, both P<0.05]; there was no statistically significant difference in the 24-hour average systolic blood pressure and average diastolic blood pressure, daytime average systolic blood pressure and average diastolic blood pressure, nighttime average systolic blood pressure and average diastolic blood pressure, baPWV, CAP and total adverse event incidence in the eplerenone group compared with the losartan group.
