What is platinib
Introduction: Platinib is a kinase inhibitor with enhanced specificity for the RET tyrosine kinase receptor (RTK) compared with other RTK classes. Platinib is an oral, once-daily, potent and highly selective RET inhibitor with strong anti-tumor activity. It selectively and effectively inhibits oncogenic RET mutations common in non-small cell lung cancer, including secondary RET mutations that may lead to treatment resistance.
Pralsetinib Indications
is indicated for the treatment of metastatic non-small cell lung cancer (NSCLC) in adult patients who have a rearrangement during transfection (RET) gene fusion as determined by an FDA-approved test. It is also indicated for the treatment of adult and pediatric patients 12 years and older with advanced or metastatic RET fusion-positive thyroid cancer who require systemic therapy and for whom radioactive iodine is not suitable. The indication for advanced or metastatic RET fusion-positive thyroid cancer is approved under accelerated approval based on overall response rate and duration of response, and continued approval for this indication may be contingent on verification and description of clinical benefit in confirmatory trials.

The mechanism of action of Platinib is mainly reflected in its selective inhibition of RET kinase. RET is a cell surface receptor tyrosine kinase that plays an important role in cell signaling. In some cancers, the RET gene becomes fused or mutated, causing it to become overactive, thereby promoting tumor growth and spread. Platinib binds to the active site of RET kinase, inhibits its kinase activity, blocks the activation of downstream signaling pathways, and thereby inhibits the proliferation and survival of tumor cells.
Medication Reference
The recommended dose of Pralsetinib is 400mg, taken once a day on an empty stomach. The doctor may adjust the dose according to the patient's needs and tolerance. Dosage for use in pediatric patients must be determined by the physician. If you miss a dose of this medicine, take it as soon as possible, but if it is almost time for your next dose, skip the missed dose and return to your regular dosing schedule without doubling your dose. If you vomit after taking one dose, do not take additional doses. Just take the next dose at your normal time the next day.
