(Vigabatrin) instruction manual

Generic name: Vigabatrin tablets

Trade name: Sabril/Vigabatrin

Other names: Vigabatrin, Sabrilex, Vigadrone

Indications: Treatment of epilepsy in which other anti-epileptic drugs are ineffective, especially partial seizures. It can also treat infantile spasms (West syndrome, West syndrome).

Mechanism of action: Vigabatrin tablets work by irreversibly inhibiting GABA aminotransferase (GABA-T) and increasing the concentration of GABA in the brain.

Usage and dosage: Vigabatrin tablets are an oral drug that is rapidly absorbed after oral administration and can reach the peak blood level 2 hours after taking it. The maximum daily dosage for adults is 2-3g. It is appropriate to start with 1g a day. Then, depending on the response after taking the medication, if the response is good, it can be increased by 0.5g every week, and finally reaches a dosage of 2-3g per day.

The initial recommended dose for children is 40 mg/kg/day. Continued medication should be judged based on the patient's weight. The recommended dose for patients weighing 10-15kg is 0.5-1g/day; the recommended dose for 15-30kg is 1-1.5g/day; the recommended dose for 30-50kg is 1.5-3g/day; and the recommended dose for over 50kg is 2-3g/day.

Contraindications: It is prohibited to use vigabatrin tablets if the patient is allergic to them and its ingredients. Pregnant women and breast-feeding patients should not use vigabatrin for treatment.

Adverse reactions: Common adverse reactions include fatigue, drowsiness, headache, dizziness, confusion and depression. Occasional aggressive behavior and psychosis (more common in patients with a history of psychosis and behavioral problems), memory loss, diplopia, weight gain, gastrointestinal disturbances, edema, and alopecia. Nystagmus, nervousness, agitation, ataxia, paresthesia, and tremor may also occur.
