The recommended oral dose of stiripentol is 50 mg/kg/day given in 2 or 3 divided doses (i.e., 16.67 mg/kg three times daily or 25 mg/kg twice daily), depending on the patient's age and weight, as shown in Table 1. If the exact dose cannot be achieved given the available strengths, round to the nearest possible dose, usually within the range of 50 mg to 150 mg of the recommended dose of 50 mg/kg/day. A combination of two stiripentol strengths can be used to achieve this dose. The maximum recommended total dose is 3,000 mg/day.

Stipentol is available in capsule and suspension forms. Stipentol capsules must be swallowed whole with a glass of water. Capsules must not be broken or opened. Stipentol oral suspension should be mixed in a glass of water (100 mL) and should be taken immediately after mixing with a meal. To ensure no medication remains in the glass, add a small amount of water (25 mL) to the drinking glass and drink the entire mixture.
As with most antiepileptic drugs, if treatment is discontinued, it should be discontinued gradually to minimize the risk of increased seizure frequency and status epilepticus. In situations where rapid discontinuation of stiripentol is medically necessary, appropriate monitoring is recommended.
If you are breastfeeding or planning to breastfeed, it is not known whether stiripentol passes into breast milk. Talk with your healthcare provider about the best way to breastfeed while you are taking stiripentol.
Tell healthcare provider about all the medicines you use, including prescription and over-the-counter medicines, vitamins, and herbal supplements. Taking stiripentol with certain other medicines can cause side effects or affect the effectiveness of stiripentol or the other medicines. Do not start or stop other medicines without telling your healthcare provider. Know the medicines you use. Keep this list and show it to your health care provider and pharmacist when you get new medicines.
