雷帕鸣(sirolimus)适应症

Rapamin (sirolimus) and FK506 are structural homologs. Although they act on the same receptor, their immune action mechanisms are different. Sirolimus, a macrolide compound, is a new and potent immunosuppressant with less toxicity discovered so far. It can prolong the survival period after transplantation, reduce the occurrence of acute rejection, and provides a new treatment alternative for transplant recipients.

What are the indications for Sirolimus?

Sirolimus Indications: Sirolimus is suitable for patients undergoing kidney transplantation to prevent organ rejection. It is recommended that Rapamune preparations be used in combination with cyclosporine and corticosteroids.

Although rapamycin is structurally homologous to TRL (Tacmlimus, FK506) and binds to the same immune-binding protein, their mechanisms of action are different. SRL inhibits the activity and proliferation of T cells and also inhibits the production of antibodies. In cells, SRL combines with the immunosuppressive protein FKBP 12 to produce an immunosuppressive complex. The SLR:FKBP 12 complex, which has no effect on calmodulin, binds to the rapamycin target site in mammalian cells and inhibits its activation. This inhibitory effect inhibits lymphokine-driven T cell proliferation and inhibits the progression from G1 to S phase in T cell evolution and differentiation.

Because rapamycin suppresses the immune system, anyone taking this drug is at increased risk for infections. This drug can slow wound healing time and increase blood cholesterol levels and blood pressure. Additionally, taking this drug increases your risk of lymphoma, skin cancer, and other types of cancer. If you notice changes in the color of skin freckles or moles, swollen lymph nodes, or unexplained weight loss, fever, or night sweats, you should tell your doctor immediately.
