Adverse reactions:

Tacrolimus: Adverse effects associated with immunosuppressants are often difficult to identify due to the severity of the patient's disease and the frequent use of multiple drugs. There is evidence that many of the adverse reactions described below are reversible and can be reduced or eliminated by reducing the dosage. The incidence of adverse reactions is lower with oral administration compared with intravenous administration. 1. Infection Just like with other immunosuppressants, patients taking Proxima may increase their susceptibility to viral, bacterial, fungal and/or protozoal infections. Existing infectious diseases may worsen. There are both systemic infections and local infections, such as abscesses and pneumonia. The risk of excessive immunosuppression may be increased if Preclosumab is used with other immunosuppressants. A comparison of patients treated with Preclosumab and cyclosporine as basic immunosuppressive therapy found that patients receiving Preclosumab.

Rapamin: Very common (incidence 10% according to International Council of Medical Organization frequency classification): lymphocele, peripheral edema, abdominal pain, diarrhea, hypokalemia, increased lactate dehydrogenase, acne, urinary tract infection; very common at higher doses: anemia, hypercholesterolemia, thrombocytopenia, hypertriglyceridemia (hyperlipidemia). Store in a cool, dry, dark environment.

Usage and dosage:

Tacrolimus: 1. Recommended starting dose for adults receiving oral proxumab after surgery: (1) For liver transplant patients, the initial oral dose should be 0.1-0.2 mg/kg based on body weight per day, taken orally in two doses, and started 6 hours after surgery. (2) For kidney transplant patients, the initial oral dose should be 0.15-0.3 mg/kg per day based on body weight, taken orally in two doses, and the medication should be started within 24 hours after surgery. 2. Rejection reaction that is ineffective in traditional immunosuppressive treatment: (1) For patients who have a rejection reaction and are ineffective in traditional immunosuppressive treatment, they should start to be treated with Proximab. The recommended starting dose is the same as the first immunosuppressive dose level. (2) When the patient is switched from cyclosporine to Preclosumab, the interval between the first doses of Preclosumab shall not exceed 24 hours.

Rapamin: Recommended for combined use with cyclosporine and corticosteroids. Take orally, once daily. It should be started as soon as possible after transplantation. For new transplant recipients, the loading dose that should be taken for the first time is three times the maintenance dose. The recommended loading dose for kidney transplant patients is 6 mg and the maintenance dose is 2 mg/day. Although the loading dose of 15 mg and the maintenance dose of 5 mg/day used in clinical trials are safe and effective, the therapeutic benefits of doses above 2 mg are unclear for kidney transplant patients. The overall safety profile of patients taking Rapamune 2 mg daily was better than that of patients taking Rapamune 5 mg daily.
