What are the side effects of Sonidegib?

Sonidegib was approved for medical use in the United States and the European Union in 2015. It is indicated for the treatment of adults with locally advanced basal cell carcinoma that has relapsed after surgery or radiotherapy, or who are not candidates for surgery or radiotherapy. Solidegib is generally administered orally and can bind to and inhibit smoothened, thereby inhibiting the activation of the Hedgehog pathway. One trial showed a higher frequency of common adverse reactions, including muscle cramps, alopecia, dysgeusia, fatigue, nausea, musculoskeletal pain, diarrhea, weight loss, decreased appetite, myalgia, abdominal pain, headache, aches, vomiting and pruritus. 34% of patients permanently discontinued Solidagib, and 20% temporarily discontinued Solidagib due to adverse reactions. At least two patients reported adverse reactions that led to discontinuation of the drug, including muscle cramps and dysgeusia, asthenia, increased lipase and nausea, fatigue, decreased appetite, hair loss, and weight loss. 18% of patients experienced serious adverse reactions.
