What are the side effects of Vandetanib?
Vandetanib (Vandetanib) is an oral multikinase inhibitor with activity against the vascular endothelial growth factor (VEGF) and epidermal growth factor (EGF) receptor families as well as RET (rearranged during transfection), BRK, TIE2 and Src kinases. It is indicated for the treatment of advanced or metastatic medullary thyroid carcinoma. The most common adverse events include diarrhea, nausea, fatigue, rash, acne, hypertension, headache, anorexia and abdominal pain.

In clinical trials, vandetanib caused QT prolongation in 14% of patients. This drug should not be used in patients with a QTc interval greater than 450 ms, nor in patients with a history of torsade de pointes, congenital long QT syndrome, bradyarrhythmias, or decompensated heart failure. Vandetanib has also been associated with photosensitivity and severe skin reactions, such as Stevens-Johnson syndrome. Patients should be advised to use sunscreen during treatment with vandetanib and for 4 months after the end of treatment. Mild to moderate skin reactions can be controlled with topical or systemic corticosteroids, oral antihistamines, or topical or systemic antibiotics. Other rare serious adverse reactions reported with vandetanib include interstitial lung disease, ischemic cerebrovascular events, serious bleeding events, heart failure, hypothyroidism, hypertension (including crises), and reversible posterior leukoencephalopathy syndrome.
