Subsidence time and treatment recommendations for side effects of sotorasib (AMG510)
Sotoracib (AMG510), as a targeted therapy for non-small cell lung cancer (NSCLC), has attracted much attention for its efficacy. However, like many medications, it can come with a range of side effects. The resolution time of these side effects varies among individuals and is affected by multiple factors such as the patient's physiological state and treatment response.
Sotorasiib works by inhibiting the KRAS G12C mutant protein, which is found in some cases of refractory non-small cell lung cancer. During treatment, some patients may experience elevated serum aminotransferases and even develop significant liver damage, which may be severe.

Clinical study data show that the most commonly reported side effects of sotorasiib include diarrhea, musculoskeletal pain, nausea, fatigue, and cough. At the same time, laboratory tests often reveal abnormalities such as lymphopenia, decreased hemoglobin, increased aspartate aminotransferase (AST) and alanine aminotransferase (ALT), decreased calcium, increased alkaline phosphatase, increased urinary protein, and decreased sodium.
About half of the patients experienced serious adverse reactions, such as pneumonia and hepatotoxicity, when using sotoraxib. Among them, significant increases in ALT and AST and diarrhea were the most common serious adverse reactions. In rare cases, patients may even die from fatal reactions such as respiratory failure and cardiac arrest. In addition, about 9% of patients had to permanently discontinue treatment due to intolerable side effects, with hepatotoxicity and drug-induced liver injury being the main reasons for discontinuation.
