Introduction: Crisaborole is a new small molecule, non-hormonal, non-steroidal anti-inflammatory phosphodiesterase 4 inhibitor, used to treat atopic dermatitis. This is a new treatment for patients with mild to moderate eczema. Research shows that criborole can be effective in treating the redness, swelling, heat, itching, rash and other symptoms caused by atopic dermatitis.

Therapeutic Mechanism

Inhibiting phosphodiesterase 4 (PDE-4) increases intracellular cyclic adenosine monophosphate (cAMP) levels. cAMP is an intracellular signaling molecule. Increased levels of cAMP can inhibit the production of inflammatory mediators. Due to the inhibition of PDE-4 activity, criborole reduces the release of pro-inflammatory cytokines, thereby reducing the inflammatory response. Criborole can quickly relieve itching symptoms in patients with AD atopic dermatitis and improve their quality of life.

Efficacy

In the clinical trials for FDA approval of criborole, researchers recruited 1,522 patients with mild to moderate atopic dermatitis and received criborole treatment or placebo ointment. In these clinical trials, after using criborole ointment twice a day for 28 days, about one-third of the participants had clear or almost clear skin.

The study found that criborrole relieved itching in more patients than placebo. On the second day of the study, 34% of patients taking criborole experienced relief from itching, compared with only 27% of those taking a placebo. At the end of the study, 57% of patients treated with criborole no longer experienced itchy skin, compared with 40% of those treated with placebo.

Dosage

Apply a thin layer of Criborole Ointment to skin as directed, usually twice daily. Dermatologists recommend applying criborole immediately after bathing and waiting 15 minutes after applying criborole before applying moisturizer. Also, wash your hands after using criborole. You do not need to wash your hands after use unless you are treating your hands. If you are not treating your hands or are using criborole on someone else, wash your hands immediately after using criborole.

Storage Conditions

Criboral should be stored at room temperature between 68°F and 77°F (20°C and 25°C), tightly closed, and out of the reach of children to prevent misuse by children.
