What can be cured? Valcyte was developed by Roche in Switzerland and launched in China in 2006. It is suitable for the treatment of patients with acquired immunodeficiency syndrome (AIDS) combined with cytomegalovirus (CMV) retinitis and the prevention of CMV infection in high-risk solid organ transplant patients.
For oral use, the recommended induction dose for the treatment of cytomegalovirus retinitis is 900 mg (two 450 mg tablets) twice daily for 21 days. Maintenance: After induction therapy, or in adult patients with inactive cytomegalovirus retinitis, the recommended dose is 900 mg orally once daily.
Adult patients should use valcyte tablets rather than valcyte oral solution. Oral solutions and tablets should be taken with food. Valcyte agent should not be broken or crushed. Before dispensing the medication to a patient, the pharmacist must prepare an oral solution (50 mg/ml) with valcyte.
Valcyte is the L-valyl ester (prodrug) of ganciclovir, which is rapidly converted into ganciclovir by esterases in the small intestine and liver after oral administration. Valcyte is a synthetic 2’-deoxyguanosine analog that inhibits herpes virus replication in vitro and in vivo.
Valcyte is teratogenic. Women should take a pregnancy test before initiating pregnancy and use effective contraception during treatment and for 30 days after treatment; men should use barrier contraception during treatment and for 90 days after treatment. Use with caution in patients with impaired renal function; dose adjustment required. Acute renal failure may occur in elderly patients with or without renal impairment; elderly patients should be used with caution and dosage adjustments based on renal function.
The most common side effects of valcyte in adults include: diarrhea, low white blood cell, red blood cell and platelet cell counts in blood tests, fever, fatigue, nausea, headache, insomnia, urinary tract infection, shaking movements (tremors), vomiting; side effects of valcyte are less frequent and generally mild. If patients experience common side effects, they can be improved and relieved by timely treatment according to doctor's instructions.
