Garampanel is a highly selective, noncompetitive AMPA-type receptor antagonist that reduces neuronal hyperexcitability associated with epileptic seizures by targeting the glutamate activity of postsynaptic membrane AMPA receptors. Currently, the 2018 AAN/AES guideline for combined treatment of refractory partial-onset epilepsy (TRAFE) in adults has a grade A recommendation. So, has Perampanel been launched in China?

On September 29, 2019, the State Food and Drug Administration approved the import license for Eisai’s new drug Parampanel. The drug is suitable for the additional treatment of adults and children aged 12 years and above with partial seizures of epilepsy (with or without secondary generalized seizures).

Perampanel is a noncompetitive alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA) receptor antagonist. Perampanel is approved as an adjunctive treatment for focal epilepsy with or without secondary generalized epilepsy in patients 12 years of age or older in more than 55 countries, including Australia, North America, Europe, Asia and Russia. Perampanel is also approved as adjunctive therapy for primary generalized tonic-clonic (PGTC) seizures in patients 12 years of age and older in North America, Europe, Asia and Russia.

The efficacy of perampanel, a partial-onset seizure treatment drug, was established in three 19-week randomized, double-blind, placebo-controlled, multicenter add-on treatment trials in adults and adolescents. Subjects had partial seizures with or without secondary generalized seizures that were not adequately controlled while receiving 1 to 3 concomitant AEDs. During the 6-week baseline period, subjects had more than 5 seizures and no more than 25 days of epilepsy-free seizures. Across the three trials, subjects' average epilepsy duration was approximately 2106 years. 85.3% and 89.1% of patients used 2-3 combined AEDs with or without vagus nerve stimulation.

Usage and dosage of Parampanel: Take once a day orally before going to bed, on an empty stomach or with food. Tablets should be swallowed whole and do not chew, crush or break. Since there are no marks on the medicine, this product cannot be opened evenly.
