Cycloserine (Cycloserine) is an antibiotic produced by microorganisms of the genus Streptomyces and is widely used to treat drug-resistant tuberculosis (MDR-TB). As an amino acid antibiotic, cycloserine can effectively inhibit the growth and reproduction of pathogenic bacteria such as tuberculosis by interfering with bacterial cell wall synthesis. 1. Inhibition of D-amino acid synthase: Cycloserine can inhibit the activity of D-amino acid synthase, thereby blocking the synthesis of peptidoglycan in the cell wall. Peptidoglycan is an important component of the bacterial cell wall. Bacteria that lose peptidoglycan will be unable to maintain the structure of their cell wall, resulting in cell wall damage.
2. Peptide transferase inhibition: Cycloserine also affects the synthesis process of bacterial cell wall by inhibiting the activity of peptidyl transferase. Peptide transferase is an important enzyme in cell wall synthesis. Cycloserine inhibits its activity, leading to interruption of bacterial cell wall synthesis, thereby damaging bacterial growth and reproduction.
Since the bacterial cell wall is crucial for bacterial survival, this mechanism of action of cycloserine makes it effective in inhibiting the growth of multiple drug-resistant tuberculosis branches, especially when other conventional anti-tuberculosis drugs fail to work. Its mechanism of action makes cycloserine an important weapon in the treatment of drug-resistant tuberculosis, especially playing a key role in combination therapy.
