Is sirolimus on the market?

The U.S. Food and Drug Administration (FDA) first approved sirolimus in 1999 for the prevention of organ rejection in kidney transplant patients aged 13 and older. In November 2000, the drug was recognized by the European Agency as an alternative to calcineurin antagonists for corticosteroid maintenance therapy. In May 2015, the FDA approved sirolimus for the treatment of patients with lymphangioleiomyomatosis.

Sirolimus, also known as rapamycin, is a macrolide antibiotic produced by Streptomyces hygroscopicus, which is isolated from soil in some areas. However, after the later discovery of its potent antitumor and immunosuppressive activities, it was extensively studied as an immunosuppressive and antitumor agent. Its primary mechanism of action is inhibition of the mammalian target of rapamycin (mTOR), a serine/threonine-specific protein kinase that regulates cell growth, proliferation, and survival. mTOR is an important therapeutic target for various diseases as it has been shown to regulate lifespan and maintain normal glucose homeostasis. Targeting mTOR has received more attention, especially in cancer, because the mTOR signaling pathway is constitutively activated in many types of human cancer
