What are the side effects of Vismodegib?

Vismodegib's side effects have been a limiting factor for the drug. All patients in the ERIVANCE trial experienced at least one adverse event. In clinical studies of vismodegib, 57% experienced mild (Grade 1) or moderate (Grade 2) adverse events. A total of 12% of patients experienced adverse events that ultimately led to discontinuation of vismodegib.

The most common (≥10%) adverse reactions with vismodegib were muscle cramps, alopecia, dysgeusia, weight loss, fatigue, nausea, diarrhea, decreased appetite, constipation, arthralgia, vomiting, and hypogeusia; grade 3 laboratory abnormalities observed were hyponatremia (4%), azotemia (2%), and hypokalemia (1%). After Vimodegib was put on the market, adverse events such as drug-induced liver injury, Stevens-Johnson syndrome (SJS), toxic epidermal necrolysis (TEN), or drug reactions with eosinophilia and systemic symptoms (DRESS) have occurred. Vimodegib can cause embryonic death or severe birth defects when used in pregnant women. It is embryotoxic, fetotoxic and teratogenic in animals. Teratogenic effects include severe midline defects, missing fingers and other irreversible malformations. Therefore verify the pregnancy status of women of reproductive potential within 7 days before starting vismodegib.
