How to take Vorinostat
The recommended dose of Vorinostat is 400 mg taken orally once daily. The response rate at this dose is 31%, and there are almost no life-threatening side effects. However, if adverse effects become intolerable, dose reduction to 300 mg once daily is warranted. Among patients treated with vorinostat at 300 mg twice daily, the overall efficacy rate was only 21%, with side effects such as pulmonary embolism and thrombocytopenia.

Vorinostat is available in 100 mg capsules approved for oral administration. A linear relationship between plasma concentration and vorinostat dose has been demonstrated with oral administration of vorinostat at doses of 200-600 mg. However,comparative analyzes of the efficacy and safety of intravenous formulations have been performed. The effectiveness of intravenous vorinostat 300-600 mg/m2, 5 days a week for 3 weeks, is similar to that of 400 mg taken orally daily. However, serious hematologic adverse reactions are quite common in patients receiving intravenous vorinostat. Second, the inconvenience of daily infusion makes the oral route more suitable for cutaneous T-cell lymphoma (CTCL) treatment. A variety of other skin-directed therapies and cytotoxic therapies should be considered in patients with systemic and progressive disease before initiating vorinostat.
The original drug of vorinostat is not currently on the market in China, and therefore cannot be included in the national medical insurance coverage. Overseas, there is an American version of the original drug, specifications100mg*120 pills per box, which may cost around 100,000 yuan (the price may fluctuate due to exchange rate effects).
