What is tisagenlecleucel?
Selevermin (Tisagenlecleucel) is an immunotherapy drug used to treat a certain type of acute lymphoblastic leukemia (ALL) in people under 25 years old. Collagen is given to tissue after other treatments have failed. Tissue leukocytes are made from white blood cells extracted from the blood drawn through veins in your body. Srifermin is available under a special program, patients must be registered in the program and understand the risks and benefits of the drug. Serivermin may also be used for purposes not listed in this Medication Guide.
Selifermin is aCD19-directed genetically modified autologous T cell immunotherapy involving genetically modified autologous T cells isolated from each individual patient. Reprogramming of patient T cells uses lentiviral vectors encoding anti-CD19 chimeric antigen receptors (CARs). The CAR consists of a murine single-chain antibody fragment (scFv) specific for CD19, followed by a CD8 hinge and transmembrane region fused to intracellular signaling domains from 4-1BB (CD137) and CD3.
These intracellular costimulatory signaling domains increaseThe expansion, long-term persistence and potential of CAR T cells; the CD3 component is critical for initiating T cell activation and anti-tumor activity, while 4-1BB enhances the expansion and persistence of tissue stem cells. After binding to CD19-expressing cells, CAR delivers signals to promote T cell expansion, activation, target cell elimination, and tissue stem cell persistence. Serifamine demonstrated efficacy in reinducing remission in patients with refractory B-cell precursor acute lymphoblastic leukemia. The sole purpose of treatment is to specifically eliminate CD19-expressing malignant and normal cells and increase the chance of remission
