Pitolisant-wakix is a first-in-class, non-controlled drug with a novel mechanism of action for the treatment of narcolepsy that sets it apart from previous treatments. It is approved for the treatment of narcolepsy types 1 and 2 and has been recognized as an orphan drug by the U.S. Food and Drug Administration (FDA) and the European Medicines Agency (EMA).

Recent advances in the treatment of narcolepsy have focused on non-hypo- and hypo-adrenergic-based therapies and immunotherapy. In Phase III trials, tilolixen tablets reduced the frequency of cataplexy episodes, reduced excessive daytime sleepiness (EDS), and improved attention levels on a sustained attention response task. Tilorisen tablets also significantly reduced hypnotic hallucinations and sleep paralysis. Additional long-term RCTs comparing tilorisen tablets with modafinil and sodium oxybate are needed to elucidate its therapeutic efficacy and potential as a first-line agent.
Tilorisen tablets have been launched in China under the trade name Hua Kesi, but they have not yet been included in medical insurance, and the specific domestic price is not yet clear. The original version of tilolixen tablets marketed overseas is sold under the trade name Wakix. It is available in two specifications: 4.45mg*30 and 17.8mg*30. The price of each box may be more than 4,000 yuan (the price may fluctuate due to the exchange rate). According to the prescription information, one box can be used for about a month (30 days), and the dosage needs to be adjusted according to the patient's specific dosage.
