How to use Eltrombopag/Eltrombopag

Eltrombopag is a hematopoietic agent used to treat patients with immune thrombocytopenia (ITP), thrombocytopenia of chronic hepatitis C, and severe aplastic anemia. It has a significant platelet-raising effect.

Eltrombopag is administered orally as tablets or oral suspension. Eltrombopag Administer at the lowest dose to achieve and maintain platelet counts ≥50, 000/mm3 to reduce bleeding risk or maintain hematologic response. Take without food or with food, low calcium content(≤50 mg). Calcium-containing foods and medications may reduce the rate and extent of eltrombopag's absorption; therefore, it is not recommended to take other medications(such as antacids) , calcium-rich foods(such as dairy products, and certain fruits and vegetables span>)or take at least 2 hours before or after supplements containing polyvalent cations(such as iron, calcium, aluminum, magnesium, selenium, zinc). If administering as an oral suspension, prepare the suspension with water only. Do not use hot water. After reconstitution, take immediately or store at 20–25°C for 30 minutes; allow 30 minutes of preparation, discard any remaining suspension. Educate patients or caregivers on the proper administration, preparation, and administration of oral suspensions. Use a new oral administration syringe to prepare each dose. Do not split, crush, or chew tablets and mix with food or liquids.

1.Forpersistent or chronicITP:For most adult and pediatric patients 6 years of age and older, start with eltrombopag at 50 mg once daily. For pediatric patients 1 to 5 years of age, initiate 25 mg of eltrombopag once daily. Patients with hepatic impairment and some patients of East Asian/Southeast Asian ancestry may require reduced dosage. Adjust to maintain platelet count greater than or equal to 50 x 109/L. Do not exceed 75mg per day.

2. For chronic hepatitis C-associated thrombocytopenia: Administer 25 mg once daily to all patients. Adjust to achieve target platelet count required to initiate antiviral therapy. Do not exceed 100mg daily.

3. For first-line severe aplastic anemia:while receiving standard immunosuppressive therapy, 2.5mg/kg (2 to 5 years old ), 7 5mg (pediatric patients 6 to 11 years old) or Eltrombopag is initiated at a dose of 150 mg once daily (in patients 12 years of age and older). Reduce initial dose in patients of East Asian/Southeast Asian ancestry. Adjust dose based on toxicity or elevated platelet count.

4. For refractory severe aplastic anemia: once daily, 50 mg each time. Reduce initial dose in patients with hepatic impairment or in patients of East Asian/Southeast Asian ancestry.
