Sotorasib (AMG510), also known as sotorasib, is being developed by Amgen and is indicated for the treatment of adult patients with locally advanced or metastatic non-small cell lung cancer (NSCLC) with a KRAS G12C mutation, as determined by a U.S. Food and Drug Administration (FDA)-approved test, who have received at least one prior systemic therapy.
Amgen's CodeBreaK clinical development program for its drug sotoraxib is designed to use the KRAS G12C mutation and address long-standing unmet medical needs in these cancers. Sotorasiib is the first KRAS 12C inhibitor to enter the clinic and is currently being studied in the largest clinical program exploring multiple combinations. Sotorasiib, a once-daily oral formulation, demonstrated rapid, deep and durable anticancer activity with a positive benefit-risk profile in patients with locally advanced and metastatic non-small cell lung cancer harboring KRAS G12C mutations. Sotoracib is also being studied in a variety of other solid tumors.

Genetic changes in the KRAS gene can produce an altered protein that causes cancer cells to grow uncontrollably. The active substance in sotorasib attaches to this altered protein in cancer cells. This prevents the protein from working, interrupting the chemical messages cancer cells need to grow and spread, and also promotes processes that lead to cancer cell death. The most common side effects of sotoraxib are diarrhea, nausea, and fatigue; the most common serious side effects are elevated levels of certain liver enzymes (which may be a sign of liver problems) and liver damage.
