Gilteritinib (brand name: Xospata) is a next-generation oral FLT3-targeted inhibitor primarily used for adult patients with FLT3 mutation-positive relapsed or refractory acute myeloid leukemia (AML). As a precision therapy, it specifically targets aberrant signaling pathways in leukemic cells, offering a new treatment option for patients harboring FLT3 mutations.

1. Precise Targeting of FLT3 Gene Abnormalities

The key advantage of gilteritinib lies in its well-defined targeting capability. It effectively inhibits the activity of FLT3 receptor tyrosine kinase, covering common mutation types such as FLT3-ITD and FLT3-TKD. FLT3 gene abnormalities drive continuous proliferation of leukemic cells, and gilteritinib blocks downstream signaling pathways to suppress abnormal cell growth, thereby enhancing therapeutic precision.

2. Oral Administration Enhances Convenience

Unlike regimens requiring intravenous infusion, gilteritinib is administered as an oral tablet once daily, regardless of food intake. This approach reduces the inconvenience of frequent infusions, facilitates long-term standardized treatment and disease management, and improves patient adherence.

3. Defined Patient Population: FLT3-Mutated Relapsed or Refractory AML

Gilteritinib is not intended for all AML patients but specifically for adults with relapsed or refractory AML who have confirmed FLT3 mutations through genetic testing. Therefore, FLT3 mutation testing must be performed before initiating therapy to determine eligibility for this precision treatment, enabling individualized medication.

4. Precision Therapy Driving AML Treatment Evolution

As AML enters the era of molecular subtype-based therapy, FLT3-targeted treatment has become a critical direction. With its precise mechanism of action, oral convenience, and well-defined indication, gilteritinib occupies a significant position in AML targeted therapy, providing more accurate and sustainable treatment options for patients with FLT3 mutations.
