kaftrio (Trikafta) indications

kaftrio (Trikafta) is a combination drug indicated for the treatment of cystic fibrosis (CF) in adults and children ‌2 years of age and older. It is only applicable to patients carrying specific CF-related gene mutations, which need to be confirmed by genetic testing (such as F508del mutation or other mutations with in vitro response).

Allergic reactions: hives, difficulty breathing, swelling of the face/lips/tongue/throat.

Visual vision changes: significant changes in vision.

‌Symptoms of liver damage‌: loss of appetite, upper right abdominal pain, fatigue, itching, dark urine, clay-colored stools, jaundice (yellowing of the skin or whites of the eyes).

1. Headache; diarrhea, abdominal pain; rash; abnormal laboratory indicators.

2. Flu-like symptoms (fever, chills, muscle aches).

3. Cold-like symptoms (nasal congestion, sinus pain, sneezing, sore throat).

Precautions for kaftrio (Trikafta)

‌Contraindicated combination drugs‌

Rifabutin, rifampicin; St. John's wort; anti-epileptic drugs (carbamazepine, phenobarbital, phenytoin).

‌Inform your doctor before taking any medicine‌

TakeBefore taking any medicine, inform your doctor of yourdrug allergies; liver or kidney disease; pregnancy or breastfeeding status.

kaftrio (Trikafta) usage and dosage

‌Dosing frequency‌

Twice a day (morning and evening, about 12 hours apart). Take 2 tablets (containing elecarpol 100mg/tezakafort 50mg/ivacaftor 75mg) orally in the morning; take 1 tablet (containing ivacaftor 150mg) in the evening.

‌Dosage requirements‌

Must be taken with foods containing fat, such as butter, peanut butter, eggs, nuts, meat, whole milk, cheese or yogurt.

Swallow the tablet whole. Do not break, chew or crush the tablet.

The granules should be mixed with 5 mL of soft food/liquid (such as puree, yogurt, applesauce, water, juice, or milk) and taken within 1 hour.

‌Children’s dosage‌

Patients under 12 years old need to adjust the dosage according to their weight and need to be re-evaluated when their weight changes.

‌Dose adjustment‌

The dosage may need to be adjusted when used in combination with other drugs. Do not change the medication plan on your own.

‌Monitoring requirements‌

Regularly test liver function.

Children need eye examinations before and after taking medication (to assess the risk of cataracts).

Handling of missed doses

Refill the dose as soon as possible, but do not take a double dose at the same time.

‌If the morning dose is missed for >6 hours‌: Take the morning dose on the same day and skip the evening dose.

‌If you miss the evening dose by more than 6 hours‌: Skip the dose and take the morning dose on time the next day.

‌All supplements must be taken with fatty foods‌.

Avoid driving or high-risk activities until the effect of the drug on reaction ability is clear.

Grapefruit products are prohibited (may cause drug interactions).
