About abrocitinib

Abrocitinib is an oral, selective JAK1 inhibitor used to treat atopic dermatitis (AD). Oral abisitinib demonstrated good efficacy and tolerability in patients with moderate to severe atopic dermatitis. The U.S. Food and Drug Administration (FDA) approved the marketing of Absitinib on January 14, 2022, and the China National Medical Products Administration (NMPA) approved the marketing of Absitinib on April 11, 2022.

The role of Absitinib

Absitinib is the first oral and effective selective JAK1 small molecule inhibitor for the treatment of psoriasis. It can regulate the expression of a variety of cytokines involved in the pathophysiological process of psoriasis, including interleukins IL-4, IL-13, IL-31, IL-22 and thymic stromal lymphopoietin (TSLP), thus exerting a therapeutic effect. Efficacy of Absitinib

Jade Mono-1 is a 12-week, multi-center, double-blind, placebo-controlled, randomized Phase III clinical trial (NCT03349060) [1], which evaluates the efficacy and safety of abisitinib in the treatment of adolescent and adult patients with moderate to severe AD for 12 weeks based on the Phase IIb clinical trial. The primary endpoints of the trial are the proportion of patients who obtained an investigator's global assessment of 0/1 at week 12 and the proportion of patients who achieved EASI-75 compared with baseline.

387 patients were included in the study and were assigned to oral abisitinib 200 mg, 100 mg and placebo according to 2:2:1. The results showed that by week 12, the proportion of oral abisitinib 200 mg and 100 mg groups receiving investigator global assessment of 0/1 was significantly higher than that of the placebo group, 44% (P<0.0001) and 24% (P =0.0037). By week 12, the EASI-75 response proportions in the oral abositinib 200 mg and 100 mg groups were significantly higher than those in the placebo group, 63% (P <0.0001) and 40% (P<0.0001) respectively. The incidence of serious adverse reactions in the oral abositinib 200 mg and 100 mg groups was very low (<4%). , but a mild decrease in platelets was observed, and the mechanism remains unclear.

This study demonstrates that once-daily oral abositinib is effective and well tolerated in adolescents and adults with moderate to severe AD, with a favorable benefit-risk profile.

Side effects of Absitinib

Common side effects of Absitinib found in clinical trials include nasopharyngitis, nausea, headache, herpes simplex, elevated blood creatinine phosphokinase, dizziness, urinary tract infection, fatigue, acne, vomiting, oropharyngeal pain, influenza, and gastroenteritis. The following are some suggestions for treatment measures:

1. Nasopharyngitis: You can use salt water gargle or nasal spray to relieve discomfort and avoid excessive nose blowing.
