[Indications]
Rucapanib (lucapanib, regapanib) is a targeted therapy. This drug is classified as a PARP (poly ADP ribose polymerase) inhibitor. It is used for patients who have received two or more chemotherapy treatments, have platinum-sensitive or platinum-resistant relapse, and have BRCA mutations. It is more suitable for platinum-resistant patients.
[Usage and Dosage]
Take 300 mg orally twice a day (about 12 hours apart). You can take it with or without food. Swallow the whole tablet without crushing or dissolving the tablet. Take the medication as directed by your doctor, and do not change the dosage or stop taking the medication without authorization. If you miss a dose, take the next dose at the usually scheduled time. Do not take extra doses to make up for a missed dose.

Common side effects (≥30%):
Elevated serum creatinine (see Kidney Problems), fatigue (including weakness), nausea, enhanced liver function tests: ALT and/or AST (see Liver Problems), vomiting, decreased lymphocyte count, anemia (low red blood cell count, constipation, cholesterol Increased appetite, loss of appetite, change in taste, diarrhea, abdominal pain
Less common side effects (10-29%):
Low platelet count, shortness of breath, dizziness, low number of neutrophils (white blood cells that fight infection), rash, fever, photosensitivity
If you experience any of the following symptoms, seek medical care immediately:
Fever 1038ºF (38ºC) ) or higher, chills (which may be a sign of infection)
Signs of an allergic reaction, such as rash, hives, itching, redness, swelling, blistering, or peeling, with or without fever, wheezing, tightness in the chest or throat, difficulty breathing or speaking, shortness of breath, new cough or worse, unusual hoarseness, or swelling of the mouth, face, lips, tongue, or throat.

[Notes]
1. Before starting treatment with rucapanib, please be sure to tell your doctor about any other drugs you are taking (including prescription drugs, over-the-counter drugs, vitamins, herbal medicines, etc.).
2. Do not take aspirin or aspirin-containing products unless specifically permitted by your doctor.
3. While taking rucapanib, do not receive any form of immunization without your doctor's permission.
4. If you are pregnant or may be pregnant, please inform your doctor before starting this treatment. Pregnancy Category D (Rucaparib may be harmful to the fetus. Women who are pregnant or pregnant must be informed of the potential harm to the fetus.)
5. For men and women: Take birth control pills and do not become pregnant while taking rucapanib (pregnancy ). It is recommended not to become pregnant for up to 6 months after the last dose of rucaparib.
6. Do not breastfeed while taking rucapanib and within 2 weeks after the final dose.
[Efficacy]
The ARIEL-2 trial evaluated the PFS of rucapanib in the treatment of platinum-sensitive relapsed patients with BRCA mutations compared with those without mutations (12.8 months vs. 5.2 months).
It is reported that the response rate of rucapanib is 66% in platinum-sensitive patients and 25% in platinum-resistant patients.
