Anamulin produced in Japan has no anti-cancer effect and mainly relieves the symptoms of cancer cachexia. Anamorelin has been approved in Japan for the treatment of cancer cachexia in patients with gastric cancer, pancreatic cancer, colorectal cancer, non-small cell lung cancer and other malignant tumors.

Anamulin does not have anti-cancer effects

It is an oral ghrelin receptor agonist used in Japan to treat cancer cachexia in malignant tumors such as pancreatic cancer, non-small cell lung cancer, gastric cancer, or colorectal cancer. Cancer cachexia can lead to weight loss and cancer-related anorexia, which can have a significant impact on patients' quality of life and prognosis.

Anamulin can promote growth hormone secretion and increase appetite through growth hormone receptor activation, thereby increasing muscle mass and weight in cancer patients. Clinical trials of anamorelin have shown that lean body mass index is significantly increased and cachexia is improved, but it does not have an anti-cancer effect on cancer.

Efficacy of Anamorelin

Anamulin is an orally available ghrelin receptor agonist. Ghrelin is a peptide mainly secreted by the stomach. It is an endogenous agonist of type 1a growth hormone receptor and regulates energy metabolism in the body by promoting growth hormone secretion, increasing appetite, and promoting lipogenesis.

Anamulin acts on ghrelin receptors in the stomach and can improve cancer cachexia by improving appetite and increasing serum insulin-like growth factor 1.

Therapeutic Effects of Anamorelin

Anamulin is a ghrelin receptor agonist that can be administered orally. A Phase 2 trial (ONO-7643-) of anamorelin for the treatment of cancer cachexia in four cancers (non-small cell lung cancer, gastric cancer, pancreatic cancer, and colorectal cancer) was approved in Japan on December 11, 2020.

For the treatment of patients with non-small cell lung cancer and cachexia, an increase in lean body mass of 1.56 kg as assessed by dual-energy X-ray absorptiometry (DXA). Another study in advanced and unresectable gastrointestinal tract (colorectal, gastric, or pancreatic cancer) showed an improvement in lean body mass of 1.89 ± 0.36 kg.

Anamorelin Dosage

The recommended adult dose of Anamorelin is 100 mg taken orally once a day on an empty stomach. However, if no effect is observed 3 weeks after starting administration, administration should be discontinued.

There is no clinical experience with administration beyond 12 weeks, and anamorelin should be used under the supervision of a physician with adequate knowledge and experience in cancer cachexia.

(Note: The above usage and dosage are from the US FDA instructions for anamorelin, which are for reference only. The specific usage and dosage should follow the doctor's advice)

Summary

Anamulin is mainly used to relieve the symptoms of cancer cachexia, rather than directly used for anti-cancer treatment. Anamorelin can help patients maintain better physical condition and improve their quality of life during anti-cancer treatment, but it is not an anti-cancer drug and does not have an anti-cancer effect.
