(Futibatinib) is a new generation of covalently bound FGFR1-4 inhibitors that has been shown to have anti-tumor activity in patients with FGFR-mutated tumors and has strong preclinical activity against acquired resistance mutations associated with ATP-competitive FGFR inhibitors. The drug was approved by the US FDA in September 2022 for the treatment of adult patients with previously treated, unresectable, locally advanced or metastatic intrahepatic cholangiocarcinoma and carrying FGFR2 fusions or other rearrangements. So, how to use forbatinib?

How to use forbatinib

20 mg (five 4 mg tablets) taken orally once daily until disease progression or unacceptable toxicity. Take forbatinib (with or without food) at approximately the same time each day, swallowing the tablet whole. Do not crush, chew, split or dissolve tablets. If the patient misses a forbatinib dose for more than 12 hours or experiences vomiting, continue with the next scheduled dose.

Forbatinib usage and dosage for special populations

1. Based on the results of an animal study and its mechanism of action, the administration of forbatinib to pregnant women can cause fetal harm or pregnancy termination.

2. Because forbatinib may cause serious adverse effects on breastfed children, it is recommended that women should not breastfeed during treatment and within 1 week after the last dose.

3. Forbatinib can cause harm to the fetus when administered to pregnant women.

4. The safety and effectiveness of forbatinib in the treatment of pediatric patients have not yet been determined.

5. Of the 103 patients treated with forbatinib in study TAS-120-101, 22% were 65 years or older. Based on the available data, no overall differences in the safety or effectiveness of forbatinib were observed between patients aged 65 years and older and younger adult patients.
