Avosidenib was developed by the French company Servier. It was approved by the US FDA for marketing in July 2018, and was officially approved by the China National Medical Products Administration (NMPA) to enter the Chinese market in February 2022.

By combining with the mutant IDH1 enzyme, it effectively inhibits the production of 2-HG and relieves the blockage of cell differentiation, allowing leukemia cells to differentiate normally into mature blood cells, thus controlling the progression of the disease.
Indications of Ivosidenib
Ivosidenib is suitable for use in adult patients diagnosed with relapsed or refractory acute myeloid leukemia (AML) carrying susceptible IDH1 mutations using well-validated detection methods.
Before using ivonib, the patient must be confirmed to have an IDH1 mutation through genetic testing.

Avosidenib usage and dosage
1. Dosage
The recommended dose is 500mg.
2. Medication time
Take orally once a day, on an empty stomach or after meals to avoid abnormal increase in blood concentration. Taking medication at the same time keeps the blood concentration of the drug at a relatively stable level.
3. Medication course
Continue medication until disease progression or unacceptable toxicity occurs. Patients who do not experience disease progression or unacceptable toxicity need to receive treatment for at least 6 months to fully observe clinical response.
4. Post-vomiting treatment
If vomiting occurs after taking the medicine, there is no need to take a supplementary dose. You only need to take the next dose according to the scheduled time.
5. Treatment after missed dose
If you miss a dose or do not take it at the scheduled time, you should take it as soon as possible. However, if it is less than 12 hours before the next scheduled dose, you do not need to take it again. You can just resume taking the medicine at the original planned time the next day. Special attention should be paid to not taking two doses within 12 hours.
Daily medication management of Avosidenib
1. Avosidenib can be taken with food or on an empty stomach.
2. Do not take it with high-fat food.
3. It is prohibited to split, crush or chew ivonib tablets.
4. Ivonib tablets should be taken orally at approximately the same time every day.
Common adverse reactions of ivosidenib
The most common adverse reactions of ivosidenib include fatigue, joint pain, diarrhea, edema, nausea, dyspnea, mucositis, rash, cough, decreased appetite, myalgia, constipation and fever, etc., with incidence rates exceeding 20%.
Proper storage of Avosidenib
Avosidenib needs to be stored in a dark, sealed, dry place with a temperature not exceeding 25°C. Exposure of medications to extremes of high or low temperatures should be avoided.
Try to store Avonim in a dry and ventilated place to avoid adverse effects on the stability of the drug. At the same time, the medication should be kept away from direct sunlight.
