Ruxolitinib tablets (Ruxolitinib) is a kinase inhibitor that inhibits Janus-associated kinases (JAKs) JAK1 and JAK2. These two kinases mediate the signaling of many cytokines and growth factors and are important for hematopoiesis and immune function. Myelofibrosis and polycythemia vera are myeloproliferative neoplasms (MPN) known to be related to dysregulation of JAK1 and JAK2 signaling. In a mouse model of JAK2V617F-positive MPN, oral administration of ruxolitinib tablets prevented splenomegaly. The JAK-STAT signaling pathway plays a role in regulating the development, proliferation, and activation of several immune cell types important in the pathogenesis of graft-versus-host disease. In mouse models, oral administration of ruxolitinib tablets was associated with reduced expression of inflammatory cytokines in colon homogenates and reduced immune cell infiltration in the colon.
