Nintedanib (Nintedanib) is a tyrosine kinase inhibitor that targets the growth factor pathway, while pirfenidone (Pirf Enidone) is an anti-inflammatory and anti-fibrotic drug that inhibits collagen synthesis and reduces fibroblast proliferation. Both drugs have been shown to slow the decline in forced vital capacity (FVC), reduce acute exacerbations, and reduce the number of hospitalizations for respiratory events. Current research shows that the two antifibrotic drugs have different side effects. Nintedanib users reported significant increases in diarrhea, vomiting, weight loss, and anorexia, whereas pirfenidone users reported primarily increased anorexia. Additionally, nintedanib was associated with nausea, while pirfenidone was associated with weight loss. Although nintedanib causes more gastrointestinal side effects, the overall burden of nintedanib side effects may be lower.
