Patients with advanced basal cell carcinoma (BCC) have very limited treatment options. Sonidegib (Sonidegib) selectively inhibits the growth of Hedgehog pathway-dependent tumors and can treat patients with locally advanced basal cell carcinoma who are not suitable for surgery or radiotherapy. These patients were randomized in a 1:2 ratio to receive 200 or 800 mg of oral solidegib daily. The primary efficacy analysis showed that 18 patients in the 200 mg group and 35 patients in the 800 mg group demonstrated objective response, corresponding to 43% and 38% of the respective categories, respectively. Among patients who took 200 and 800 mg solidegib, 93% and 80% of patients respectively had their disease controlled. The most common adverse events were muscle cramps, alopecia, dysgeusia, nausea, elevated blood creatine kinase, andfatigue. In contrast, patients who took 200 mg of Solidagib experienced fewer adverse events than those who took 800 mg of Solidagib. Therefore, the benefits of using the 200 mg dose of Solidagib outweigh the associated risks, and it can be concluded that it would be prudent to choose a lower intensity dose.
