Efficacy of Romiplostim

In a 24-week Phase III trial, Romiplostim was significantly better than placebo in achieving the primary endpoint of a protocol-defined durable platelet response in non-splenectomized or splenectomized adult patients with chronic immune thrombocytopenic purpura. In clinical studies, treatment with romigrastim resulted in a dose-dependent increase in platelet counts.

After a single subcutaneous injection of 1-10mcg/kgromigrastim in patients with immune thrombocytopenia (ITP), within 2-3 weeks, the peak platelet count is 1.3-14.9 times higher than the baseline platelet count. Among 8 patients with ITP who received 1 mcg/kg of romiplostim for 6 weeks, 7 had platelet counts greater than 50 × 109/L. In a clinical study, after a single intravenous dose of 10 mcg/kg romiplostim in healthy adults, the peak platelet count increased 4.7-7.3 times compared with the baseline value (mean: 5.8 times). Results from population modeling and simulations indicate that a single 10 mcg/kg subcutaneous dose of romigrastim will have a clinically relevant impact on the incidence and duration of severe thrombocytopenia in patients acutely exposed to myelosuppressive doses of radiation.
