What is Ponatinib?

Ponatinib (Ponatinib) is a prescription medicine used to treat adults with: chronic myeloid leukemia (CML), including chronic phase, who are intolerant to or no longer benefit from treatment with at least 2 prior kinase inhibitor drugs; accelerated phase or Blast phase chronic myelogenous leukemia, or Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ALL), who cannot receive any other kinase inhibitor drugs; Chronic phase, accelerated phase, or blast phase CML with a specific type of abnormal gene (T315I positive), or T315I-positive Ph+ ALL.

Ponatinib is not indicated for the treatment of patients with newly diagnosed chronic phase chronic myelogenous leukemia, and it is not known whether ponatinib is safe and effective in children. Ponatinib is a multi-target kinase inhibitor. Its main cellular target is the Bcr-Abl tyrosine kinase protein, which is constitutively active and promotes the progression of CML. The protein comes from the fused Bcr and Abl genes, commonly known as the Philadelphia chromosome. Ponatinibis unique in that it is particularly suitable for the treatment of drug-resistant CML because it inhibits the tyrosine kinase activity of Abl and T315I mutant kinases.
