The effects and efficacy of Palbociclib

Oral palbociclib (Palbociclib) is the first highly selective cyclin-dependent kinase 4 and 6 inhibitor (i.e., CDK4/6 inhibitor). It is indicated for the treatment of women with HR-positive, HER2-negative advanced or metastatic breast cancer in combination with an aromatase inhibitor as initial endocrine therapy and with fulvestrant (with or without a luteinizing hormone-releasing hormone agonist) in women who have received prior endocrine therapy.

In clinical trials, palbociclib combined with letrozole as initial endocrine therapy for postmenopausal women (PALOMA-1 and PALOMA-2), or combined with fulvestrant for premenopausal, perimenopausal, or postmenopausal women with disease progression after endocrine therapy (PALOMA-3), significantly prolonged progression-free survival (PFS) and improved clinical benefit response (CBR) rates. Neutropenia was the most commonly reported adverse event of any grade and grade ≥3 and was usually manageable by delaying, interrupting, or reducing the palbociclib dose without routine use of growth factors and without compromising efficacy.

The original drug of palbociclib has been launched in China and has entered the scope of Class B medical insurance, but reimbursement is limited to patients who meet the indications. The price of each box may be more than more than 4,000 yuan. The price of palbociclib's original drug, 125mg*21 capsules, per box sold overseas may be more than RMB 10,000 (the price may fluctuate due to exchange rates). There are also generic palbociclib drugs produced in other countries.
