Why should you take opicapone at night?

Opicapone increases the bioavailability of levodopa and has significant, long-lasting, and sustained inhibition of catechol oxygen methyltransferase when administered as a once-daily evening oral regimen and/or 1 hour apart from levodopa treatment. Opicapen and placebo were well tolerated and similar. By taking it in the evening or at least one hour apart from levodopa, the amount of levodopa available after a dose is increased, but any interaction between the two drugs is avoided during the levodopa absorption phase.

Results from two largePhase III trials show that within a week of starting treatment with Opicapone, it begins to work and that patients who received Opicapone 25 mg per day spent an average of 61 minutes less resting, on average, and those who received Opicapone 50 mg per day, compared with 23 minutes on placebo. Further improvements were observed during the second and third weeks of treatment, after which treatment effects stabilized. At the end of the 14-15 week study, significant reductions in off time were observed of 93 minutes for recipients of Opicapone 25 mg and 120 minutes for recipients of Opicapone 50 mg, compared with a reduction of 55 minutes for placebo recipients.

Opicapone has not yet been launched in the country, so it is not covered by medical insurance.
