Can Obinutuzumab cure follicular lymphoma?

A randomized, open-label, multicenter trial based on demonstration of improvement in progression-free survival (PFS) in patients with follicular lymphoma (FL) who did not respond or progressed during or within 6 months of treatment with a rituximab-containing regimen. The trial compared 6 cycles of obinutuzumab plus bendamustine, followed by 2 years of obinutuzumab monotherapy and 6 cycles of bendamustine.

The treatment regimen for otuzumab consists of 1,000 mg intravenously on days 1, 8, and 15 of cycle 1; on day 1 of cycles 2-6 (28-day cycle); and then every two months for two years. Bendamustine is administered as an intravenous infusion of 90 mg/m2 on Days 1 and 2 of Cycles 1-6. The efficacy of 321 patients with follicular lymphoma was evaluated and randomly divided into the otuzumab plus bendamustine group or the bendamustine group. The median age is 63 years. Patients had received a median of 2 prior therapies. Independent review assessed that the median PFS was 13.8 months in the bendamustine group and was not reached in the otuzumab plus bendamustine group [HR 0.48 (95% CI: 0.34-0.68).
