Alemtuzumab’s mechanism of action
Alemtuzumab is a monoclonal antibody drug whose mechanism of action is related to the regulation of the immune system and immunosuppression. Its main function is to affect T lymphocytes and B lymphocytes in the immune system by specifically binding to the CD52 antigen. The following are key steps in alemtuzumab’s mechanism of action:
1.CD52Antigen binding: CD52Antigen is a protein present on the surface of a variety of immune system cells, including T lymphocytes, B lymphocytes and natural killer cells. Alemtuzumab binds to CD52antigen with high specificity to form an antibody-antigen complex.
2.Immune system modulation: Once alemtuzumab binds toCD52antigen, it triggers an immune system response. Specifically, it causes cell-related toxic reactions, including cell-mediated cytotoxicity (Cell-Mediated Cytotoxicity, CMC) and antibody-mediated cytotoxicity (Antibody-Dependent Cellular Cytotoxicity, ADCC).

3.Lymphopenia: The mechanism of action of alemtuzumab causes the destruction or reduction ofCD52antigen-positive lymphocytes. This includes T cells and B cells, which play important roles in autoimmune diseases and some blood cancers.
4.Immune system“Reset”: After lymphocytes are consumed, the immune system will gradually recover and “Reset”. This may reduce abnormal immune responses such as those seen in autoimmune diseases such as multiple sclerosis.
5.Therapeutic effect: In leukemia types such as chronic lymphocytic leukemia (CLL) and some autoimmune diseases, the mechanism of action of alemtuzumab can help reduce disease symptoms, improve patients' quality of life, and even achieve long-term remission.
