The average response time to venetoclax in chronic lymphocytic leukemia was 0.8 months, with duration of response ranging from 2.4 to 52.4 months.

After several oral doses after meals, maximum plasma concentrations of venetoclax are reached 5 to 8 hours after the third dose. The steady-state AUC (area under the curve) of venetoclax increased proportionally over the dose range of 150-800 mg. After a low-fat meal, at the 400 mg once-daily dose label, the mean steady-state Cmax of venetoclax was 2.1 ± 1.1 μg/mL and the AUC0-24 was 32.8 ± 16.9 μg·h/mL. Compared with the fasting state, venetoclax exposure increased 3.4-fold when consuming a low-fat meal and 5.2-fold when consuming a high-fat meal. When comparing low fat to high fat, both Cmax and AUC increased by 50% when high fat meals were consumed.
