How to take Vemurafenib
Vemurafenib (Vemurafenib) is indicated for the treatment of patients with unresectable or metastatic melanoma who have a BRAF V600E mutation detected by a U.S. Food and Drug Administration (FDA)-approved test, and is also indicated for the treatment of patients with Erdheim-Chester disease who have a BRAF V600 mutation. However, it is not suitable for treating patients with wild-type BRAF melanoma.
Before starting treatment with vemurafenib in patients with melanoma, doctors will confirm the presence of the BRAF V600E mutation in melanoma specimens. The recommended dose of vemurafenib is 960 mg (four 240 mg tablets) orally every 12 hours, with treatment continued until disease progression or unacceptable toxicity. Missed doses can be taken up to 4 hours before the next dose. If adverse reactions occur after treatment with vemurafenib, the medication should be adjusted, interrupted or stopped under the guidance of a doctor. If toxicity occurs for the first time, it can be reduced to 720 mg twice a day. If the toxicity recurs for the second time and is highly toxic, the dose needs to be further reduced. It is recommended to reduce the dose to 480 mg twice a day. If the dose is lower than this dose, the treatment will be ineffective.
