How long does it take for Vemurafenib to take effect?
Vemurafenib has been approved to treat patients with BRAFV600E-positive unresectable or metastatic melanoma based on demonstrated survival benefit in a randomized phase III study. The currently approved dosing regimen of vemurafenib is 960 mg orally twice daily until disease progression or unacceptable toxicity occurs. Vemurafenib is well tolerated, and the most common side effects include skin reactions, photosensitivity, headache, and joint pain.
The response rate of vemurafenib was evaluated in a clinical study of vemurafenib 960 mg twice daily in patients with metastatic melanoma whose tumors tested positive for the BRAF V600E mutation. Among 32 patients, 81% achieved an objective response, the majority of which was partial. Tumor regression was observed in the patient population, even in those with low-risk characteristics, such as visceral organ involvement or elevated lactate dehydrogenase levels. The onset of response seems to be early, with symptoms improving within 1 or 2 weeks of starting treatment. Duration of response ranged from 2 to 18 months, with median progression-free survival of 7 months or longer.
