What is Temsirolimus?

The active ingredient in Temsirolimus ( Temsirolimus) blocks a protein called the mammalian target of rapamycin (mTOR). In the body, temsirolimus attaches to a protein inside cells, forming a "complex." This complex then blocks mTOR. Because mTOR is involved in the control of cell division, temsirolimus can prevent cancer cells from dividing, thereby slowing the growth and spread of cancer.

In an international three-arm Phase III study, temsirolimus, interferon-alpha, or their combination were compared in 626 previously untreated patients with poor prognosis. The median overall survival in the temsirolimus group (10.9 months) was significantly improved compared with the interferon-alpha group (7.3 months) and the combination group (8.4 months). After cancer patients took a single dose of 25mg temsirolimus, the average temsirolimus Cmax in whole blood was 585ng/mL, and the average AUC in blood was 1627ng*h/mL. Sirolimus exposure increased in a less than dose-proportional manner over the dose range of 1 mg to 25 mg, whereas sirolimus exposure increased in a dose-proportional manner. After a single intravenous injection of 25 mg in cancer patients, the AUC of sirolimus is 2.7 times that of temsirolimus, mainly due to the longer half-life of sirolimus.

The original drug temsirolimus has not yet been marketed in China, and therefore cannot be included in domestic medical insurance. The original temsirolimus drug marketed overseas has a Turkish version and a European version. The ingredients of the two are basically the same. The price of each box of 30mg/1.2mL may be more than 10,000 yuan, and the price of 25mg/mL may be around 3,000 yuan per box (the price may fluctuate due to exchange rates).
