Are temsirolimus and sirolimus the same?
Temsirolimus (Temsirolimus) is a prodrug of sirolimus, both of which are pharmacologically active inhibitors of the mammalian target of rapamycin. Mammalian target of rapamycin (mTOR) is a serine/threonine kinase involved in many important cellular functions, including cell growth, proliferation, angiogenesis, and cell survival. Temsirolimus and its major metabolite sirolimus form complexes with fk-506-binding protein, an intracellular immunophilin, and these complexes exert anti-angiogenic and anti-tumor activities by inhibiting mTOR.

Temsirolimus was approved for the treatment of advanced renal cell carcinoma (RCC) after clinical trials showed that intravenous temsirolimus prolonged overall survival in patients with advanced renal cell carcinoma (RCC) compared with interferon α treatment. Sirolimus, also known as rapamycin, is a macrolide compound used to coat coronary stents to prevent organ transplant rejection. It can also treat a rare lung disease called lymphangioleiomyomatosis. It has immunosuppressive functions in the body and is particularly useful in preventing kidney transplant rejection.
The original drug temsirolimus has not yet been marketed in China, and therefore cannot be included in domestic medical insurance. The original temsirolimus drug marketed overseas has a Turkish version and a European version. The ingredients of the two are basically the same. The price of each box of 30mg/1.2mL may be more than 10,000 yuan, and the price of 25mg/mL may be around 3,000 yuan per box (the price may fluctuate due to exchange rates). There is currently no generic version of temsirolimus available on the market. For more drug information and specific prices, please consult a medical consultant.
