What are the precautions for taking Tazemetostat?

In clinical studies of Tazemetostat (Tazemetostat) treatment, warnings and precautions such as secondary malignant tumors and embryo-fetal toxicity have emerged.

1. Secondary malignant tumors: After receiving tazerestat treatment, the risk of secondary malignant tumors increases. In a clinical trial of 729 adults taking tazerestat 800 mg twice daily, myelodysplastic syndrome (MDS) or acute myeloid leukemia (AML) occurred in 0.7% of patients. A pediatric patient developed T-cell lymphoblastic lymphoma (T-LBL). Physicians should monitor patients over the long term for the development of secondary malignancies.

2. Embryo-fetal toxicity: According to the results of animal studies and its mechanism of action, tazetostat taken by pregnant women can cause fetal damage. Administration of tazerestat to pregnant rats and rabbits during organogenesis resulted in a dose-dependent increase in skeletal developmental abnormalities in both species, starting from maternal exposure that was approximately 1.5 times the adult exposure (area under the plasma concentration-time curve [AUC0-45h]) at a dose of 800 mg twice daily. Doctors should inform pregnant women of the potential risks to the fetus. Tazerestatmay interfere with the action of hormonal contraceptives (pills, patches, rings, implants, or injections), so these should not be used as the only method of contraception during treatment.

The original drug of tazerestat is not yet available in the domestic market and is therefore not included in medical insurance. Tazerestat US version of the original drug, specification200mg*240 tablets, which is marketed overseas, may cost around 200,000 yuan per bottle (the price may fluctuate due to exchange rates), which is very expensive.
