How long does it take for Tazemetostat to work?

The onset of action of tazemetostat (Tazemetostat) varies between individuals. Generally, some initial signs of efficacy, such as tumor shrinkage or cessation of growth, may be observed within a few weeks of starting treatment. However, it usually takes several months to achieve optimal results. Tazerestat is available as an oral tablet. The recommended dose is 800 mg taken orally twice daily with or without food until disease progression or unacceptable toxicity occurs. If a dose is missed or vomiting occurs after taking tazerestat, do not take the additional dose but continue with the next scheduled dose.

Tazerestat is a potent, selective, orally bioavailable small molecule inhibitor of EZH2 enzyme activity. EPZ-6438 specifically induces apoptosis and differentiation in SMARCB1-deficient MRT cells. Systemic exposure to tazerestat was approximately dose-proportional (0.25 to 2 times the approved recommended dose) over the dose range of 200 mg to 1,600 mg of tazerestat administered twice daily. Steady state was reached on day 15 after oral administration of tazerestat 800 mg twice daily. The average steady-state peak plasma concentration (Cmax) was 829ng/mL, and the AUC0-12h was 3340 (49%) NGH/mL. Tazerestat exhibits time-dependent pharmacokinetics (PK). The average cumulative ratio (measured by AUC) was 0.58. High-fat, high-calorie (approximately 800 to 1,000 calories) meals did not have a significant effect on tazemitos exposure.

The original drug of tazerestat is not yet available in the domestic market and is therefore not included in medical insurance. Tazerestat US version of the original drug, specification200mg*240 tablets, which is marketed overseas, may cost around 200,000 yuan per bottle (the price may fluctuate due to exchange rates), which is very expensive.
