How to take Omarigliptin

Omarigliptin (Omarigliptin) The recommended dose is 25 mg taken orally once weekly. If the patient misses a dose, take it as soon as he remembers. Then take your next dose according to your normal dosing schedule. Never take a double dose to make up for a missed dose. If a patient experiences adverse reactions while taking ologliptin, the doctor may adjust the drug dose based on the severity of the condition, such as reducing the dose to 12.5 mg orally once a week in patients with severe kidney impairment.

A semi-mechanistic 2-compartment model fully describes the PK of ologliptin with linear unbound clearance and concentration dependence on binding of ologliptin to the DPP-4 enzyme in the central and peripheral compartments. Key covariates for ologliptin PK include decreased unbound clearance in renal impairment. The direct effect sigmoid maximal inhibitory potency model adequately describes the relationship between ologliptin plasma concentrations and DPP-4 inhibition. These models support the current Japanese labeling indication that the approved dose of ologliptin 25 mg once weekly is halved in patients with severe renal impairment and patients with end-stage renal disease. Additionally, if a patient misses a dose, the next dose of ologliptin should be taken as soon as possible from memory, up to and including the day before the next scheduled dose. No other clinically important covariates were identified.

The original drug Ologliptin is not marketed in the country and is therefore not included in medical insurance.
