Sotorasib’s mechanism of action
The mechanism of action of Sotorasib (Sotorasib) is mainly focused on combating the KRAS gene G12C mutation in non-small cell lung cancer (NSCLC). KRASThe gene plays a key role in cell growth and differentiation, and when the KRASgene undergoes the G12C mutation, it will cause the cell's growth signaling pathway to be out of control and promote the abnormal proliferation of cancer cells.
Sotoracib is aKRAS G12Cinhibitor that directly interferes with this specific gene variant. Its mechanism of action can be summarized into the following key steps:
1.Specific binding: Sotoraxib binds to the KRAS G12C mutation site to form a drug - protein complex. This binding is highly specific, primarily targeting the G12C mutant form of the KRAS gene, but does not have the same affinity for other types of KRAS mutations.

2.Inhibition ofKRASactivity: The combination of drugs causes the activity ofKRAS G12C mutation to be inhibited, blocking its normal growth signaling. This effect directly weakens the abnormal proliferation ability of cancer cells, thereby inhibiting tumor growth.
4.Blocking the signaling pathway: KRAS G12CMutation is usually related to the RAS-MAPK signaling pathway, and the effect of sotorasiib blocks this signaling pathway. This helps slow down the rate of cell proliferation, thereby slowing the progression of cancer.
5.Inducing apoptosis: Studies have shown that the effects of sotoraxib may also reduce the survival ability of cancer cells by inducing apoptosis, a process of self-destruction in cancer cells.
The original drug of sotorasibu has not yet been launched in China, so patients cannot purchase it domestically and need to purchase sotorasibu through overseas channels. The price of the original version of sotorasibu in Hong Kong, China, with the specifications of 120mg*240 tablets, is around 8 million, and the price of the European version of the original drug with the same specifications is around 4 million, which is relatively expensive. The cheaper ones are the generic drugs marketed abroad. For example, the price of generic drugs in Laos and Bangladesh is around two to three thousand yuan. The price is relatively cheap, and the ingredients of foreign generic drugs are basically the same as the original drugs.
